You can't in the main story mode unless you play as player 2, 3, or 4. In Guest Star, however, you can play as the Dream Friends once you beat one campaign with a normal enemy. And in Ultimate Choice I think you can choose any characters you have unlocked

Don't forget you can play as them in certain sections of Heroes in Another Dimension!
